Studies on Streptomyces phage II. Host range and species differentiation.

The phage 514-3 of Streptomyces griseus described in the preceding paper (Gilmour et al., 1959) has from the time of its isolation from soil, showed a high infectivity for a large number of diverse Streptomyces species. The application of pure-lining procedures involving transfers from plaques and continued propagation on the homologous host S. griseus strain 3475 has not altered the observed broad host range of the phage. These observations, although indicative of the host infectivity spectrum, were regarded as qualitative in nature since only mycelium lysis was used as the index of phage infection. It is the purpose of the present paper to present host specificity data based on more precise measurements. The latter included detection of lysogenic strains, and plating efficiency tests. Several streptomyces groups emerged, based on the plating efficiency of developed phage preparations in selected host cultures. The principle of host controlled variation of phage effected by passage through new hosts (Cragie and Yen, 1938; Luria and Human, 1952; Bertani and Weigle, 1953) was followed in the development of the various phage lysates. Similarities and differences among the test Streptomyces species were observed by appropriate cross infection patterns.
